The hotel was built in 1585 by Bernardino Contarini. The hotel's name comes from Saint Daniel of Padua, who is the patron saint of the Hospital of San Giorgio Maggiore. The building was originally intended to be used as an orphanage and later became a hospital for children with physical and mental disabilities. In 1884, it was converted into a hotel and became known as Hotel Danieli Venice.

Throughout its history, Hotel Danieli has hosted many notable guests including Queen Victoria and Prince Albert II of Monaco, King Edward VII of Great Britain, Napoleon III of France, Emperor Hirohito of Japan, Countess Sophia Tolstoy and her son Alexei Alexandrovich Tolstoy (who later became Tsar Alexander II), Sir Winston Churchill and his wife Clementine Churchill (who stayed at the hotel during World War II), Prince Rainier III of Monaco and his wife Grace Kelly (who stayed at the hotel during their honeymoon).
